This girl is holding a house party and invited along a girl who I am interested in. Of course we have spoken before and defo feel she is interested in me. The problem is I feel I will get more out of girls who I don't know rather than girls who I have already spoken to because what do I say and how do I game her?

I don't understand your question exactly, if you're interested in this girl or want to meet other girls.
If you're interested in this girl then you should talk to her some but also talk to other people at the party, including guys and girls. Then come back to her again later. It's actually in your favor that there are a lot of people you can talk to.
